摘要如下： Statins reduce risk for cardiovascular diseases but their role in dementia prevention is unclear. We conducted a prospective cohort study of statin use and incident dementia and Alzheimer's disease (AD) in 2356 cognitively intact persons aged 65 + in the Seattle area using a pharmacy database. A total of 312 participants developed dementia (AD=168) during the study period. The adjusted hazard ratios (HR) with statin use were 1.19 (95% CI 0.82 - 1.75)for dementia and 0.82 (CI 0.46 - 1.46) for probable AD. A subgroup analysis of participants with at least one APOE-ε4 allele who entered the study before age 80 produced an adjusted HR of 0.33 (CI 0.10 - 1.04) for AD. The findings from recent cohort studies and large randomized placebo-controlled trials do not support that statin use reduces overall risk for dementia.
