Two decades ago, studies in hemophilic animals demonstrated that liver-directed gene transfer could induce immune tolerance to therapeutic transgene products. These early studies addressed a major concern in the emerging field of gene replacement therapy for genetic diseases and potentially opened up gene transfer to other applications such as autoimmune diseases.1 Subsequent studies further demonstrated that liver-directed gene transfer could even eliminate antibodies in animals pre-immune to the therapeutic protein resulting from prior protein exposure.
