This colorimetric micromethod for the determination of inorganic phosphate, avoiding deproteinisation, was examined for linearity and reproducibility. We compared the method with the methods of S. Raabe (Rec. Trav. Chim., 74 (1955) 561) and D. B. Zilversmit and A. K. Davis (J. Lab. Clin. Med., 35 (1950) 155). The influence of deproteinisation on the results was studied and deproteinisation proved to be superfluous. In addition the normal range of inorganic phosphate in serum from a mixed, adult population from 613 blood donors was estimated. The normal values are between 0.55 and 1.33 mmol P/1. Clinical experience with this method was obtained from children during their stay in hospital (premature babies, babies, infants). This very sensitive and simple method is excellently suited for the routine determination of inorganic phosphate in serum and urine. Inorganic phosphorus as low as 0.05 μg can be determined with good precision. The linearity is satisfying between 0.03 and 6.46 mmol P/1. The values obtained by other methods are higher than that obtained with the Malachite green micromethod. We believe that phosphate groups are hydrolysed from organic compounds during deproteinisation.
