Renal protection of Ginsenosides Rg1 and its mechanism in D-galactose-induced subacute aging mice

Objective To observe the renal protection of Ginsenoside Rg1 in D-galactose-induced subacute aging mice and to investigate the mecheanism. Methods The 36 Kunming mice of clean grade were randomly divided into three groups,normal group( n = 12),model group( n = 12) and experimental group( n = 12). Model group and the experimental group were given intraperitoneal injection of D-galactose in order to make subacute aging mice,the experimental group injected with Ginsenosides Rg1 20 mg ·( kg · d)-1,for 12 weeks. The normal group was injected with normal saline. The contents of malondialdehyde( MDA),superoxide dismutase( SOD),and lipofuscin in renal tissue were measured after 12 weeks by Kit. The expression and distribution of transforming growth factor-β1( TGFβ1) and fibronectin( FN) in renal tissue were detected by immunohistochemical assay. And the level of TGFβ1 was investigated by Western-blot. Results Lipofuscin and MDA in the model group were markedly increased while the activities of SOD decreased significantly in comparison with those in the control group( P 0. 05). The lipofuscin and MDA decreased significantly in the experimental group( P 0. 05),the activities of SOD were higher than those in the model group,and has significant difference( P 0. 05).There were no obvious difference of lipofuscin,MDA and SOD activity between the experimental group and the normal group. Compared withthe control group,the expression of TGFβ1,FN in model group was significantly increased( P 0. 05). The expression of TGFβ1,FN in the experimental group was significantly lower than those in the model group( P 0. 05). The renal expression of TGFβ1 was increased remarkably in model group,and was reduced significantly in the experimental group( P 0. 05). Conclusion Ginsenoside Rg1 has protective effect on D-galactose-induced subacute aging mice.
