摘要
Adapting methods from traditional high performance liquid chromatography (HPLC) to ultra-HPLC (UHPLC) has been the focus of many laboratories in recent years. Transferring methods, namely isocratic and gradient methods, from UHPLC to traditional LC platforms potentially addresses the analytical method transfer limitations of UHPLC within the industry and allows labs to execute Quality by Design robustness studies using UHPLC methods and then to scale the conditions back to more commonly available HPLC platforms. Because of this diversity of liquid chromatographic platforms available across the industry, there has been much interest in how to transfer HPLC methods to the UHPLC platform, as well as in returning to HPLC from a UHPLC platform. This chapter focuses on the adaptation of chromatographic method conditions from one platform to the other.
