摘要
Numerous advancements in drug delivery systems are currently underway to mitigate the gross toxicity of cytotoxic drugs and enhance their therapeutic efficacy and selectivity. The implementation of site-specific drug delivery to tumors is of utmost significance in order to minimize undesirable adverse effects. Liposomes, as nanoscale vesicles, exhibit substantial potential as nanomedicines in cancer therapy. The progressions in liposomes advanced to the development of pH-sensitive liposomes that mainly exploit the tumour microenvironment. In this review, the rationale, formulation consideration, application and the formulation associated challenges in the scale-up manufacturing, patents, along-with the regulatory consideration are summarized. The regulatory agencies are working in this drug delivery system to set and provide the guidance for industry in scale-up and manufacturing. Additionally, it summarises the recent research work done of pH-sensitive liposomes over the past few years for the management of cancer, which is not covered so far. Distinctive overview and recent case studies on pH-sensitive liposomes gives an understanding on potential of this drug delivery in cancer therapy.
