The present study was undertaken with the objectives of sequence characterization and identifi cation of polymorphisms within exons 6, 7 and 13 and their fl anking intronic regions in the bubaline lactoferrin gene. Single strand conformation polymorphism analysis revealed a total of eight different variants in three investigated regions of the lactoferrin gene. The locus LtfE6 revealed two SSCP variants with the frequency of 0.75 and 0.25 respectively while four different variants, viz. LtfE7-A, LtfE7-B, LtfE7-C and LtfE7-D, were observed at the locus LtfE7 with the frequency of 0.11, 0.25, 0.47 and 0.17, respectively. The locus LtfE13 revealed two SSCP variants with the frequencies of 0.92 and 0.08, respectively. Comparison of nucleotide sequences of different regions of the buffalo lactoferrin gene with that of taurine cattle revealed a total of 26 point mutations, 13 of which were found to be in coding DNA region. Conceptualized translation of nucleotide sequence revealed two amino acid changes, viz. serine to threonine and arginine to alanine within exons 7 and 13, respectively. Analysis of nucleotide sequences of different SSCP variants showed 100 per cent homology within each locus indicating polymorphism revealed by SSCP could be independent of sequence variation.
