Objective To study the effect of Cu Ling Pian (GLP) on serum levels of osteoprotegerin (OPG), receptor activator of nuclear factor-κB ligand (RANKL), carboxyterminal propeptide of type Ⅰ procollagen (PICP), and other related cytokines (IL-6, TNF-α, IL-4) in castrated rats. Methods Rats were randomly divided into sham-operated group, model group, high dosage GLP group, middle dosage GLP group and low dosage GLP group. Osteoporosis (OP) rat model was established by castration. A month after operation, GLP groups were fed with GLP. Three months after feeding, serum levels of OPG, RANKL, PICP, IL-6, TNF-α, and IL-4 were evaluated by ELISA. Results Compared to the model group, serum levels of OPG and PICP were increased significantly (P<0.05 and P<0.01, respectively), while levels of RANKL, IL-6 and TNF-α were decreased significantly in the three GLP groups (P<0.01, P<0.01 and P<0.05, respectively). And in the GLP of high and middle dosage group, serum levels of IL-4 were increased significantly (P<0.05). Conclusion The prevention and therapeutic effect of GLP on OP may be via the up-regulation of OPG and down regulation of RANKL, IL-6 and TNF-α. This would affect the function of osteoclast and inhibit bone resorption. On the other hand, up-regulating of PICP probably may result in the increase of type Ⅰ procollagen, which finally affected the bone tenacity and intensity.
