Publisher Summary Enzyme kinetics is the study of the rates of reactions involving enzymes. This includes the study not only of overall reactions catalyzed by enzymes but also of the individual component steps if they can be measured separately. The topic can be divided into steady-state kinetics and rapid-reaction kinetics. Steady-state measurements are usually made over a time scale of minutes; they involve small molar concentrations of the enzyme under study and relatively much higher concentrations of substrates. What is measured is the gradual disappearance of a substrate or accumulation of a product as enzyme molecules undergo multiple catalytic cycles. Rapid-reaction studies, on the other hand, typically involve much higher molar concentrations of enzyme, similar to the concentrations of substrates. They are usually on a time scale of 1–1000 msec and concentrate on the anatomy of a single turnover. In rapid-reaction studies, it is often possible to observe the changing concentrations of individual enzyme–substrate complexes as well as substrates and products.
