In this research,in vitro culture method was developed for obtaining Huperzia serrata thallus which producets Huperzine A,an anti-Alzheimer's disease(AD) drug candidate. The semi-woody branches of wild mature plant of huperzia serrata from Lushan were chosen to be the test material. The test results indicate that the suitable medium to induce thallus was 6,7-V + IAA0. 5 mg /L,and its differentiation rate was 50%. Also,MS minimal medium without any hormone was established as the optimal subculture medium. Given sugar amount of 20 g /L in this subculture medium,the relative increasing rate of thallus was 1362. 6% after 55 days,namely 14. 6 folds biomass of inoculation. Through Thin-layer Chromatography and High Performance Liquid Chromatography detections,the production of active ingredients Huperzine-A by the obtained thallus has been confirmed.
