Objective:To extract high quality genomic DNA from Lonicera Japonica Thunb.dry buds with silica gel. Methods: Two different methods,conventional CTAB method and modified CTAB method,were applied to isolate genomic DNA from dry leaves of Lonicera Japonica Thunb.The DNA obtained was analyzed by the means of agarose gel electrophoresis,ultraviolet spectrophotometric method and enzymatic digestion.Results:Higher purity DNA was obtained by modified CTAB method and it can be digested by restriction enzyme EcoRⅠcompletely. The DNA extracted from dry buds had no obvious differences with the DNA from fresh buds in quality and got higher yield. Conclusion: The modified CTAB method was suitable for the extraction of genomic DNA from Lonicera Japonica Thunb. dry leaves.
