Probiotic supplements are products that are widely used for the modulation of human microbiota. The current study aims to define two formulas of a probiotic product (named PROEXO) in order to determine its influence on simulated microbiota using the GIS2 three-chamber colonic fermentation system. Polysaccharides synthesized using lactic acid bacteria (LAB) strains were lyophilized and used to simulate the effect of prebiotics on favorable strains. The results showed that polysaccharides induce increases in the LAB strains in the ascending segment of the colon. The effect is similar to those seen in other oligosaccharides, which were able to increase the cell proliferation of favorable strains from the gut. The ability of LAB strains to synthesize polysaccharides represents a major advantage of the PROEXO product because it functions as both a probiotic and prebiotic. After the utilization of two PROEXO probiotic formulas an increase of favorable strains was determined, especially in the case of v.1, in the transverse and descending colon segments. The administration also determined a significant decrese of staphylococci and clostridia.
