摘要
Objective To investigate the effects of hypoxia-reoxygenation(H-R) injury on the expressions of PI3K,Akt and hypoxia inducible factor-1α(HIF-1α) mRNA in rat cardiac microvascular endothelial cells(MMECs).Methods The MMECs of SD rats were cultured in 96-well plates(100μl/hole) or in dishes of 6cm in diameter(2ml/dish) with the density of 1×106/ml,which then were randomly divided into 2 groups of H-R(n=6,treated with hypoxia for 2 hours and reoxygenation for 2 hours) and N(n=6,normal control).The cell vitality,apoptotic rate and expressions of PI3K,Akt and HIF-1α mRNA were detected at the end of reoxygenation.ResultsCompared with group N,the cell vitality was decreased,the apoptotic rate increased and the expressions of Akt and HIF-1α mRNA up-regulated in group H-R(P0.05 or P0.01).ConclusionH-R of SD rats myocardium increases the apoptotic rate and reduces the vitality and up-regulates the expressions of Akt and HIF-1α mRNA,which is one of the mechanisms of MMECs H-R injury.
