Whole cell extracts, purified organelles, and organelle subtractions from organotrophically grownEuglena gracilis Klebs strain Z and the derived aplastidic mutant, PR-4, were assayed for the synthesis of the phylloquinone precursors,o-succinylbenzoate and 1,4-dihydroxy-2-naphthoate. The enzymes responsible for the synthesis ofo-succinylbenzoate from isochorismate and 2-ketoglutarate were localized in the cytosol and linked to the presence and development of the photosynthetic apparatus. Tentative evidence was obtained for the synthesis of 1,4-dihydroxy-2-naphthoate fromo-succinylbenzoate; the enzymes appeared to be associated with the chloroplasts and synthesis occurred at the chloroplast envelope membrane.
