A Method for production and purification of Human IL_2

A method for production and purification of human interkin 2(IL2) was described. Mono- nuclear cells of peripheral blood from donors wers cultured in RPMI1640 with 10%FCS for 4 days at 37℃.Then the cells were deposited by centrifugation and resuspended in RPMII64O with 1%FCS and 2μg/ml PHA, followed by mixing with an equal vocun of B. Lymphoblastoid cell line (Daudi, Imkl101) suspension. After 18-24h of incubation, the supernatant designated as crude IL2 was collected. The Purification scheme of this crude IL2 involved precipitation with ammonium sulphate, C-100 gel filtration and Blue-Sepharose chrcmotography. A 1000- fold purification of IL2 from crude supernatant was obtained.
