The goldenberry (Physalis peruviana L.) is one the most cultivated and edible fruit-bearing species of the Physalis genus. Several repositories harbor large collections of the species and related taxa. This germplasm has been characterized through morphological and agronomical variables, cytogenetics, and different types of molecular markers. Furthermore, genome-wide association studies for fruit quality, yield, and the resistance response to Fusarium oxysporum f. sp. Physali (Foph), have been used in combination with single-nucleotide polymorphisms for candidate gene discovery. The list of candidate genes related to Foph has also been enriched through transcriptomics and validated through differential gene expression and quantitative PCR. The above represents part of the molecular diversity and prebreeding toolbox of the species. The goal of this chapter is to present a comprehensive overview of goldenberry research involving germplasm characterization along with genome and transcriptome studies to leverage for the species' conservation and breeding potential.
