These studies have uncovered common as well as disease-specific lymphoma drivers that represent promising therapeutic targets; however, significant barriers to clinical translation still remain. In this chapter, we review the role of NGS in identifying molecular drivers of lymphomagenesis. We survey computational and experimental methods for identifying and validating lymphoma driver genes. We review recurring genetic themes in B- and T-cell lymphomas that reflect normal lymphocyte biology and summarize the genetic landscapes of the most common lymphoma types with their characteristic driver genes and pathways. We discuss various approaches for creating genomic subgroups in diffuse large B-cell lymphoma and the challenges associated with each approach. Finally, we summarize the opportunities and challenges for leveraging genomic subgroups or biomarkers to inform the course of treatment.
