Abstract: Various neurodegenerative disorders like Alzheimer’s disease, Parkinson’s disease, Huntington disease, and amyotrophic lateral sclerosis are becoming a major issue in global life. Several treatment options are available, but the major problem of all these approaches is the presence of the highly protective blood-brain barrier (BBB) which limits the possible drug transportation towards CNS. BBB particularly allows some specific components like small sized lipophilic agents through the capillary endothelial membrane. The major challenge to delivering the drug into CNS is the presence of the highly protective BBB; thus, currently, a nanocarrier-based drug delivery system has been approved for an effective and safe approach. This review has depicted a clear view about the route of nanocarrier based drug administration, the reason behind choosing nanocarriers, and what are the several nanocarriers (such as lipoplex, polyplex, nano-micelles, dendrimers, exomes, nanogels, etc) that used to treat the mentioned brain disorders and potential risk, future aspects.
