BACKGROUND: Allografting by using alginate-polylysine-alginate (APA) microencapsulated cells has been confirmed an effective way of avoiding immunological rejection induced by receptor. OBJECTIVE: To observe the effect of transplanted APA-microencapsulated ovarian cells on the biological function of adrenal gland. DESIGN, TIME AND SETTING: A randomized controlled animal experiment was performed at the Reproductive Medicine Center, Capital Medical University from March 2007 to September 2008. MATERIALS: Ovarian cells were separated from Wistar rats and APA-microencapsulated. METHODS: A total of 40 female Wistar rats were randomly divided into 4 groups. In ovariectomy group, ovaries on both sides were extirpated at sterile. In implantation group, the APA-microencapsulated ovarian cells were implanted into abdomen of the ovariectomized rats. In estrogen replacement therapy group, diethylstilbestrol was injected into abdomen of ovariectomized rats (0.2 mg/kg/3 d). No treatment was performed in the control group. MAIN OUTCOME MEASURES: At 30 days after intervention, radioimmunity assay was used to detect estrogen changes in the serum, morphology of adrenal gland was observed, and SP method was used to detect proliferating cell nuclear antigen (PCNA) expression. RESULTS:① Radioimmunity assay indicated that estrogen level in the normal control group, estrogen replacement therapy group and the implantation group was significantly higher than the ovariectomy group, but there was no remarkable difference in the normal control group, implantation group, and estrogen replacement therapy group. ② The width and the relative value of reticular zone, glomerulosa and fascicular zone in adrenal gland in the ovariectomy group become higher than the other three group. PCNA-positive cells in the adrenal gland of ovariectomy group were significantly higher than implantation group and estrogen replacement therapy group. CONCLUSION: APA-microencapsulated ovarian cells can continuously synthesize and secrete estradiol (E2), which is used to correct morphological changes following ovariectomy. The therapeutic effect of APA-microencapsulated ovarian cells is highly closed to that of estrogen replacement treatment.
