Glaucoma is a group of neurodegenerative diseases characterized by loss of retinal ganglion cells and visual field defects and is one of the major causes of irreversible blindness worldwide. Primary open-angle glaucoma (POAG) is one of the classifications of glaucoma. Oxidative stress in trabecular reticulated cells is one of the possible mechanisms of the development of glaucoma. At present, there is still a lack of effective methods to treat glaucoma. Ghrelin is characterized by its wide distribution and high potency and has anti-inflammatory, antioxidant, and anti-apoptotic effects, which may be beneficial in the treatment of glaucoma. In this study, we investigated whether ghrelin can protect human trabecular meshwork cells (HTMCs) from oxidative damage induced by hydrogen peroxide (H2O2), as well as the possible mechanism of action. CCK8 and flow cytometry results revealed that treatment of HTMCs with ghrelin showed a dose-dependent protective effect against H2O2-induced damage. Ghrelin significantly decreased the rate of apoptosis and levels of reactive oxygen species (ROS) and malondialdehyde (MDA) and increased the level of superoxide dismutase (SOD) and catalase (CAT) in HTMCs. The difference was statistically significant compared with the H2O2 group. Ghrelin activated Nrf2/HO-1/NQO-1 signaling pathways and decreased HIF-1α level in H2O2-injured HTMCs as shown on qPCR and Western blot. In conclusion, ghrelin can protect HTMCs from oxidative damage induced by H2O2 and reduce apoptosis in HTMCs, which can be a new approach to treating POAG. The underlying therapeutic mechanism may be related to Nrf2/HO-1/NQO-1 signaling pathways and HIF-1α. Le glaucome est une maladie neurodégénérative caractérisée par une perte progressive des cellules ganglionnaires de la rétine et des défauts du champ visuel, et est l'une des principales causes de cécité irréversible dans le monde. Le glaucome primaire à angle ouvert (GPAO) est le principal type de glaucome. Le stress oxydant dans les cellules trabéculaires humaines (CTH) est l'un des mécanismes possibles du développement du GPAO. À l'heure actuelle, il y a encore un manque de moyens efficaces pour traiter les GPAO. La ghréline se caractérise par sa large distribution et son potentiel élevé, elle a des effets anti-inflammatoires, antioxydants et anti-apoptose, et peut être bénéfique pour le traitement du glaucome. Dans cette étude, nous avons examiné si la ghréline peut protéger les CTH contre les dommages oxydatifs induits par le peroxyde d'hydrogène (H2O2) et son mécanisme d'action possible. Les résultats du CCK8 (Kit de comptage cellulaire-8) et de la cytométrie en flux ont montré que les CTH traitées avec de la ghréline présentaient des effets protecteurs dose-dépendants contre les dommages induits par le H2O2. La ghréline a considérablement réduit les taux d'apoptose ainsi que les niveaux d'espèces réactives de l'oxygène (ROS) et de malondialdéhyde (MDA), et les niveaux accrus de superoxyde dysmutase (SOD) de catalase (CAT) dans les CTH. La différence est statistiquement significative par rapport au groupe H2O2. Les résultats de qPCR et de Western blot ont montré que la ghréline activait la voie de signalisation de Nrf2/HO-1/NQO-1 et réduisait les niveaux de HIF-1α dans le modèle de dommages aux CTH induits par H2O2. La ghréline pourrait donc protéger les CTH contre les dommages oxydatifs induits par le H2O2, réduire le taux d'apoptose des CTH et peut constituer une nouvelle piste thérapeutique du GPAO, intéressant les voies de signalisation Nrf2/HO-1/NQO-1 et HIF-1α.
