Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases with functions in tissue morphogenesis and remodeling. An expanded view of their substrates and functions reveals them as important signaling mediators in diverse immune and inflammatory processes. MMP dysregulation contributes to many diseases – for some of which, individual MMPs can offer targets for therapy. Here, we overview current knowledge of MMP catalysis, regulation, and inhibition from a structural and mechanistic perspective, with implications for therapeutic development.
