To investigate the effects of maternal hypergalycemia on the development of early mouse embryo by the technique of in vitro fertilization,6~8 week old ICR female mice models of diabetes mellitus(DM) were established by two intraperitoneal injections of Streptozotocin(STZ),and the control group was set up.Blood of the mice were collected by posterior orbital venous plexus for the test of 14 biochemical indexes.The DM mice were superovulated by the intraperitoneal injection of PMSG and hCG,and oocytes were fertilized with sperms of normal male mice in vitro,and cultivated in HTF medium for 24 hours.The results showed that blood glucose levels of DM mice increased,significantly(P0.001),blood glucose levels in 76.6% of treated mice were above 9.5 mmol/L.Compared with the control mice,Alanine aminotransferase,creatinine,low density lipoprotein,cholinesterase and glycated hemoglobin in DM mice were significantly increased(P0.01).The fertilization rate of the oocytes of DM female mice with the sperms of normal male mice in HTF medium was lower than these of normal female mice.There was a tendency of a higher of blood glucose level with a lower of fertilization rate.We have a conclusion that hyperglycemia in maternal mice still has some adverse effects on early embryo development although in vitro fertilization technology can improve the rate of fertilization.
