Background: Lung cancer is a leading cause of cancer-related deaths worldwide. MiRNAs are small noncoding RNAs that have important roles in the regulation of gene expression in both functions as tumor suppressors or oncogenes. Single nucleotide polymorphisms (SNP) inside the miRNA genomic region contribute to cancer susceptibility and development. Previous studies reported that the SNP Rs11614913 in miR196a is correlated with hepatic and renal cancer. Aim: In this study we aimed to evaluate the association of the rs11614913 polymorphism in miR196a with the risk of non-small cells lungs cancer (NSCLC) in an Iranian population. Methods: 49 confirmed NSCLC patients and 50 healthy control referred to the Masih Daneshvari Hospital, Tehran-Iran were recruited between April 2015-September 2017. Allele frequencies for SNP Rs11614913 from genomic isolated DNA from PBMC was evaluated by polymerase chain reaction - restriction fragment length polymorphism (PCR-RFLP) and the PCR product digested with the restriction enzyme Taal to identify the mir-196 C/T polymorphism. Differences in genotype distribution between NSCLC and healthy subjects were analyzed using a Chi-square test. P values <0.05 were considered statistically significant. Results: Our data indicates that the rs11614913 polymorphism in miR-196a is significantly increased in NSCLC patients (CC vs. TT+TC: OR= 2.43, 95%CI= 1.06 – 5.54, P= 0.034). Conclusion: The current study shows a significant correlation between rs11614913 polymorphism in miR-196a with increased the risk of NSCLC in an Iranian population with NSCLC. Additional larger cohort studies are required to confirm this data.
