A VAMP-associated protein OsVAP1-3 assists the endoplasmic reticulum exit of OsNRAMP5. The S313F variation in OsNRAMP5LAA 23 weakens its interaction with OsVAP1-3 and causes partial retention of OsNRAMP5LAA 24 in the endoplasmic 25 reticulum, consequently diminishing Cd and Mn uptake in rice. Pyramiding the weak allele OsNRAMP5LAA and the strong allele OsHMA3LAA 26 substantially reduces grain-Cd 27 concentrations without compromising Mn-SOD activity and stress tolerance in rice
