Total RNA was isolated from cultured cells and enteroids and colonoids, and homogenized colon tissues, using an RNA isolation kit (Qiagen, Valencia, CA) and was treated with RNase-free DNase. Reverse transcription was performed using the High Capacity cDNA Reverse Transcription kit and the 7300 RT-PCR System (Applied Biosystems, Foster City, CA). The data were analyzed using Sequence Detection System V1.4.0 software. Primers, Setd1b (Mm00616971), Tgfb1 (Mm01178820), TNF (Mm00443259), and Ifng (Mm99999071) were purchased from Applied Biosystems. The relative abundance of β-actin mRNA was used to normalize levels of the mRNAs of interest. All complementary DNA samples were analyzed in triplicate.
