OBJECTIVE To establish a RP-HPLC method for the determination of the content of galangin in Alpinia officinarum Hance.METHODS An optimum HPLC conditions with binary mobile phase system were obtained.The samples were separated on a Shim-pack VP-ODS column(2.0 mm×250 mm,5 μm).The mobile phase consisted of 0.1%HAc aqueous solution as phase A and 15% acetonitrile+40% methanol+45% tetrafuran as phase B.Gradient elution program was as followed: 0-20 min,39%-61%B;20-30 min,61%~100%B;30-40 min,100%B.The flow rate was 0.2 mL·min-1,column temperature was 35 ℃ and UV was at 280 nm.RESULTS The chromatographic resolution of glangin was above 1.5.A good linearity was obtained with the correlation coefficients ranged of 0.020-0.64 g·L-1(r=0.999 8).The mean recovery was 100.7%,RSD was 2.82%(n=6).CONCLUSION The method is reliable,accurate and specific,which can be used for the quality control of Alpinia officinarum Hance and its products.
