Objective: To observe the effect of triterpenoidal saponins from Achyranthes bidentata Bl.on osteoclast differentiation,and to elucidate their potential mechanism on antiosteoporosis. Methods: The bone marrow cells were cultured in the presence of PTH to form tartrate-resistant acid phosphatase(TRAP)-positive multinucleated cells,then triterpenoidal saponin samples at the concentration of 2,20,and 200 μM were added to test the effect on osteocalst differentiation. Results: Seven triterpenoidal saponins showed inhibitory activity on osteoclast differentiation,among which chikusetsusaponin Iva and momordin Ib showed more potent inhibitory activity,and the inhibitory activity were reversible.Conclusion: Triterpenoidal saponins from Achyranthes bidentata Bl.can inhibit osteoclast formation,indicating that they can be used as bone resorption inhibitor to treat osteoporosis.
