OBJECTIVE To prepare diclofenac diethylamine(DDEA)hydrogel patch and study the effect of penetration enhancers on percutaneous absorption of DDEA in vitro.METHODS The DDEA hydrogel patch was prepared with hydrophilic and biocompatible polymer materials.Isolated rat abdominal skin was taken as a model using modified Franz diffusion technique.The DDEA content in the receiver at different times was determined by HPLC.The cumulative amount and the permeation kinetic parameters of DDEA were calculated.RESULTS Transdermal enhancers could increase percutaneous absorption of DDEA.Menthol had the most obvious permeation-enhancing effect.There was a positively related dose-effect relationship of menthol from 1% to 5% on increasing the penetration of DDEA.When the amount of menthol was 5%,the steady-state penetrating rate was 18.121 μg·cm-2·h-1,and the enhancement ratio was 5.45.CONCLUSION The menthol could be used as transdermal enhancer for DDEA hydrogel patch.
