Objective To study relationship between histopathology of osteoarthritis and metabolism of chondrocyte cultured in vitro. Provided theoretically foundation for treating osteoarthritis. Methods Osteoarthritic model was induced in rabbits left knee, making it extension by immobilization, right as normal reference. Isolated rabbit′ s chondrocytes aquired from healthy and osteoarthritic cartilage were cultured in vitro. DNA,CSRPG and Collagen in the cultured liquids were measured to estimate chondrocyte′ s ability of proliferating and synthesis ground substance, The severity of osteoarthritis was graded according to a modified histopathologic score originally discribed by Mankin. Results DNA. CSRPG and Collagen in cultured liquids were position correlated with severity of disease through Mankin′s 10-11, however. they are rapidly declined after Mankin′s 10-11,both matrix synthesis and cell proliferation were strongly reduced in the severe grades of osteoarthritis. Conclusions The phenotype of rabbit′s articular chondrocytes come from healthy and different grades of osteoarthritis was remained in Vitro, indicating a regulatory mechanism in the osteoarthritic chondrocyte itself probability regulating matabolism. The alter of the osteoarthritic cell matabolism in Vitro was directly related to the histopathologic grade .
