Circular dichroism (CD) has been extensively used to characterize proteins, especially protein structural changes, protein–ligand interactions, and structural response to external stimuli. CD signals result from the interaction between chromophores that may possess intrinsic chirality or are kept in a chiral environment. In this chapter we discuss how this technique is used to determine the secondary structure of peptides.
