The safety and contraceptive effectiveness of quinestrol-quingestanol (Q-Q) in oil filled capsules taken every 3 or every 4 weeks was studied in 159 patients for 1457 cycles. All but 2 of the women had previously been pregnant. The true pregnancy rate was 2.5 per 100 woman years. Withdrawal bleeding began 3-14 days after Q-Q administration. Those menstrual disorders that occurred were more frequent during the first 3 cycles. Endometrial biopsies up to 10 days after Q-Q administration were progressively secretory. After 12 days endometrium was considered proliferative. Side effects noted were gastrointestinal disorders neuropsychiatric manifestations i.e. nervousness depression headache dizziness and fatigue mucorrhea mastalgia and skin disorders. Laboratory tests performed in all patients were within normal limits.
