摘要
There are hundreds of combinations of linker, payload and antibody format in producing an antibody-drug conjugate (ADC) with a proper safety and efficacy. During drug development, it is crucial for researchers to fully understand the metabolites and distribution of antibody-drug conjugates in circulation in human body, the data of which could be used to further optimize the drug design and to further improve the efficacy in clinical applications. In this chapter, we introduced the process of ADCs' distribution and metabolization, with following discussions about the factors that influence the pharmacokinetic process of ADCs when these molecules are administered into blood system. At last, we introduced some commonly used detection methods that enable researchers to trace the metabolites after metabolization happening.
