2-aminopyrimidines can be classified as privileged structures that cross over into multiple therapeutic indications. 2-Aminopyrimidines represent an important class of bioactive molecules due to the presence of strategic points in the core framework, where judicious manipulation would introduce structural diversity. There are a number of 2-aminopyrimidines small-molecule FDA-approved drugs available in the market that showcase these privileged structures. This chapter discusses the applications of 2-aminopyrimidines in the treatment of cardiovascular, central nervous system, inflammatory, metabolic and oncology diseases. Direct substitution of 2-methylsulfonyl- and 2-halopyrimidines with primary and secondary amines via SNAR reactions is the preferred method of choice in the preparation of substituted 2-aminopyrimidines.
