Nanogels are a promising drug delivery system as they can hold drugs and macromolecules and can also respond to external stimuli, which makes them effective tools for various biomedical applications. They are nano-size dispersions of the three-dimensional crosslinked network of hydrophilic polymers, which possess high stability and tunability. The delivery of drugs through encapsulation into nanogel can prevent their degradation and also reduce the side effects. It enhances the bioavailability of drug molecules by prolonging their half-life and decreases fast clearance. The targeting capability and blood circulation time of nanogels can be modified by surface functionalization. This review provides an overview of different preparation methods and release mechanisms of nanogel, along with its application in various diseases such as cancer, cardiovascular diseases, etc. In order to resolve problems with conventional therapies, such as non-targeted delivery, side effects, low stability, etc., nanogels are a promising new drug delivery technology. This review will help researchers develop better nanogels with desired characteristics and enhance the usefulness of nanogels for various biomedical and theranostic applications.
