AbstractThe rapid development of metabolic engineering and synthetic biology has recently attracted great attention to exploring many non-model microbial species. Pichia pastoris and Pichia kudriavzevii are the two representative Pichia species, which possess uniquely desired biochemical, metabolic, and physiological features favoring large-scale industrial production. This review begins with illustrating synthetic biology parts and tools that have been developed for manipulating these two species, followed by summarizing their applications in biomanufacturing as hosts to produce recombinant proteins, bulk chemicals, and natural products, and concludes on the challenges and potential strategies for advancing them for broader biotechnological applications.
